Update on the Availability of a New iPSC Stock

RESEARCHES

Regarding the QHJI01s04 cell line provided by our Foundation, we would like to inform you that a new cell stock has been established and is now available for provision, following our announcement dated January 14, 2025.

Overview

In our announcement dated January 14, 2025, our Foundation reported that a deletion mutation in the EP300 gene had been detected in some differentiated cell samples derived from the QHJI01s04 cell line (hereinafter referred to as “this cell line”).

Subsequently, at our Facility for iPS Cell Therapy (FiT), we performed subcloning using the original parental cell line and selected clones in which the EP300 mutation was not detected by ddPCR analysis. These selected clones were subsequently used to establish a new cell stock. We have conducted comprehensive testing and evaluation of this cell stock in accordance with our Foundation’s quality control standards and have confirmed to meet the applicable quality requirements.

Provision of QHJI01s04 Cell Stock

For those wishing to obtain this cell line, in addition to the conventional SCS (Secondary Cell Stock; 24-1), we will begin offering a cell stock (24-2) that has been screened for the EP300 mutation using ddPCR.
Depending on your intended application and your institution’s risk assessment, you may select one of the following options:

  • 24-1:QHJI01s04(QHJI01s04 SCS(P12))
  • 24-2:QHJI01s04-B(QHJI01s04 EP300 mutation-negative Cell Stock(P19))

Please note that standard whole-genome analysis of the conventional SCS (24-1) has not detected any mutations in genes listed in cancer-related gene databases, including EP300.

Furthermore, since the announcement dated January 14, 2025, no safety concerns or adverse events have been reported that are considered attributable to the EP300 mutation in nonclinical studies or clinical trials using differentiated cells derived from this cell line.

Our Foundation will continue to collect and evaluate quality information regarding iPS cell stocks, including this cell line, and will share the necessary information in a timely manner. If you have any questions or would like additional materials, please contact us at the address below.
